Abstract:
In this experiment, sodium dithionite was used as a reducing agent in a Sephadex G-25 fine gel column to convert methemoglobin to hemoglobin. Qualitative indication of reduction was seen as the brown colored methemoglobin passed through the column and exited as a red eluant. Quantitative comparison was shown using a spectrophotometer. The experimental absorbance spectrum obtained for HbO2 was shown to agree within 3.3% of the literature absorbance spectrum.
