Abstract:
The design of this project is to carry out a precise determination of the material balance of a sample material placed in a column by means of a chromatograph. The material balance would result in a calibration to directly determine the mass of sample coming off the column. From the mass of sample collected, the fraction of sample remaining in the column can be found. For three successful hemoglobin runs, the masses of protein exiting column were 2.522 + 0.083 mg, 2.402 + 0.069 mg, and 2.422 + 0.080 mg. The mass loaded onto the column was calculated to be 2.764 + 0.440 mg. Given the high degree of propagated error in the loaded mass (15.9%), the residual mass of protein in the column could not be determined. This result suggests that steps need to be taken to improve the mass determination of the initial protein loaded, since that contributed the greatest amount of error in determining the mass of protein exited.
