My Research:

Over the past 10 years, I have been focusing on development of next generation point-of-care diagnostic system in the following aspects:

Development of Next Generation Point-of-Care Diagnostic System Based on Microfluidic Chip and Nucleic Acid Isothermal Amplification.

Key results include: (1) Designed a point-of-care device and firstly developed specific Loop-Mediated Isothermal Amplification (LAMP) primer set for molecular detection of Zika Virus. (2) Established a two-stage isothermal amplification process, dubbed RAMP, achieving high level of multiplexing (16, more is feasible) with order of magnitude higher sensitivity than single stage assays, high speed (<40 min), and high tolerance to inhibitors. Designed a corresponding chip for automatically distributing first-stage products to second-stage reactor. (3) Created a disposable microfluidic cassette for molecular detection of schistosome infections.

Application Research of Novel Materials (Conjugated Polymers and Nanoparticles) in Next Generation Point-of-Care Diagnostic System.

My contributions are: (1) Developed a series of methods for gene mutation detection using water-soluble cationic conjugated polymers, which have profound clinic applications. (2) Established a new system for sensitive and selective detection of circulating tumor cells by combining Aptamer-immuno-PCR and signal amplification of conjugated polymers. (3) Discovered a new self-aggregation phenomenon of single stranded (ss) DNA-modified gold nanoparticles (ssDNA-GNPs), which was used for DNA visualized detection. In this work, I also clarified the mechanism of non-crosslinking aggregation of gold nanoparticles that is confounded in academia for a long time.
